Pelle (PLL) may be the kinase that phosphorylates both Cactus (CACT) and the Dorsal/Dorsal-related immunity factor (DL/DIF) homodimer but the biochemical evidence for this is currently inconclusive. DL/DIF homodimer is phosphorylated while associated with CACT. DL is phosphorylated on Ser317 while DIF is very likely to be phosphorylated at Ser348 (inferred from DL). CACT is phosphorylated in its N-terminal half within the first 125 residues. In this region, there are two IKK target-like motifs, involving Ser74 and Ser78 in one, and Ser116 in the other. However, it is not clear which of these residues need to be phosphorylated.
