In humans, the Cactus (CACT) orthologue IkappaB is polyubiquitinated by the beta-TrCP E3 ubiquitin ligase containing complex. This marks it for degradation by the 26S proteosome. It is believed that CACT is degraded in a similar manner, however, a Drosophila E3 ubiquitin ligase that acts in the Toll pathway has yet to be identified. The obvious candidate, the closest beta-TrCP orthologue Slimb (SLMB) has produced contradictory evidence on whether it participates in the Toll pathway (Spencer et al., 1999; Leulier et al., 2003).
