Based on studies with the oncogenic fusion protein NPM-ALK, SRC (and potentially other SRC-family members) binds to activated ALK receptor. Binding depends on tyrosine residue 1358 and ALK kinase activity, identifying pY1358 as the SRC-binding site (Cussac et al, 2004). SRC signaling downstream of ALK contributes to cellular proliferation (Cussac et al, 2004; reviewed in Chiarle et al, 2008; Palmer et al, 2009).
