Production of type I IFN genes in response to Toll-like receptor 3 (TLR3) and TLR4 ligands is mediated by TANK-binding kinase 1 (TBK1) or I-kappa-B kinase epsilon (IKKε, IKBKE), which phosphorylate IFN regulatory factor 3 (IRF3) and IRF7. The activity of TBK1 and/or IRF3, IRF7 is regulated by multiple mechanisms including post-translational modifications, protein-protein interactions, and protein degradation (Zhao W et al., 2013; Runde AP et al., 2022).
