Activated (phosphorylated) PDPK1 (PDK1) phosphorylates PKN2 (PRK2) on threonine-816 located in the activation loop (Balendran et al. 2000, Dettori et al. 2009, Jin et al. 2021). Threonine-958 of PKN2 may also be phosphorylated (Dettori et al. 2009). A hydrophobic motif in the C-terminal region of PKN2 interacts with the kinase domain of PDPK1 (Balendran et al. 2000, Dettori et al. 2009). Laminar flow stress on endothelial cells activates PDPK1 to phosphorylate PKN2, which then phosphorylates AKT1 on threonine-308 (Jin et al. 2021).
