ADAM17 cleaves Notch1 at S2
By expressing membrane bound mouse Notch1 construct NdeltaE lacking amino acids 21-1703, it was shown that the cleavage at the S2 site precedes cleavage at the S3 site performed by the gamma-secretase complex. Human ADAM17 isolated from HeLa cells was found to be capable of cleaving NdeltaE at S2.
