Gamma secretase complex cleaves Cntn1-actived Notch1
By expressing myc-tagged full length recombinant mouse Notch1 in rat immature oligodendrocyte cell line, it was determined that activation of Notch1 by recombinant mouse Cntn1 is followed by accumulation of Nicd1 in the nucleus. The cleavage of Notch1 involved S3 cleavage, as determined by exogenous expression of S3 cleavage site mutants of mouse myc-tagged Notch1, and required activity of the gamma-secretase complex.
