p-Creb, Ascl1, Tcf12, p300 bind the vgf promoter
Vgf expression in rat PC12 cells is governed in part by the binding of p-S133 Creb, Ascl1 (also known as Mash1) and Tcf12 (also known as Heb) to cognate binding sites in the proximal promoter. Tcf1, which acts as a Vgf repressor in non-neuronal tissue, binds to the E box and may heterodimerize with Ascl to weakly transactivate expression (Di Rocco et al, 1997; Mandolesi et al, 2002). p-Creb binding to a CRE site upstream of the transcriptional start site is required for NGF-responsive transcription, and may be aided by binding of an unidentified factor to an adjacent CCAAT site (D'Arcangelo et al, 1996; Mandolesi et al, 2002). The CRE- -binding complexes is also associated with p300 (Mandolesi et al, 2002).
