28S subunit binds MITF2:GTP, mRNA, and formylMet-tRNA
The 28S subunit in a complex with MTIF3 (IF-3Mt, IF3mt) binds mRNA and, at some point, MTIF2:GTP (IF-2Mt:GTP, IF2mt:GTP) (Liao and Spremulli 1991, Ma and Spremulli 1996, Koc and Spremulli 2002, Christian et al. 2010, Yassin et al. 2011). If an initiation codon is present at the 5' end of the mRNA then N-formylmethionyl-tRNA also binds and a stable, productive initiation complex results (Christian and Spremulli 2010). If no initiation codon is present the 28S subunit slides along the mRNA and then dissociates. MTIF2 in mitochondria appears to perform the functions of both IF1 and IF2 in bacteria (Gaur et al. 2008, Yassin et al. 2011).
