Levels of Vrille (VRI) protein rise in parallel with the rise in vri mRNA transcription. As VRI accumulates in the nucleus between midday and dusk, it binds to the VRI/PAR domain protein-1 (PDP1) binding sites (V/P boxes) to repress transcription of its target genes, Clk and cry. As VRI levels rise, the superior VRI/PDP1 ratio means it competitively replaces PDP1 as the main binder to Clk and cry genes.
