The substitution of aspartic acid residue with tyrosine at position 560 in the iSH2 domain of PIK3R1 relieves inhibition of PI3K catalytic subunit PIK3CA and enables constitutive activity of PI3K complex that contains PIK3R1 D560Y mutant (Jaiswal et al. 2009).
