In transient transfection assays performed in human hepatoma HepG2 cells, NR1H3 (LXR alpha) was found to interact with the NCOA1 (SRC1) and NCOR cofactors to regulate the UGT1A3 gene promoter (Verreault M et al. 2006). Unliganded LXR:RXR actively suppresses transcription by recruiting the co-repressor NCOR complex. Agonist binding induces dissociation of the co-repressor, which results in moderate stimulation of transcription and later recruits co-activators such as NCOA1 leading to maximal stimulation of transcription.
