Co-immunoprecipitation assays in influenza A virus (IAV)-infected human leukemia monocytic THP1 cell, as well as NS1-transfected human embryonic kidney (HEK293) cells showed that viral NS1 interacts with mixed-lineage kinase domain-like protein (MLKL) in an RNA-dependent manner (Gaba A et al. 2019). The second brace helix of MLKL is responsible for interacting with NS1 (Gaba A et al. 2019). The interaction of NS1 with MLKL is thought to increased MLKL membrane translocation. Moreover, the MLKL:NS1 interaction enhanced NLRP3 inflammasome activation and increases IL-1β processing and secretion (Gaba A et al. 2019).
