MITF-M binds to the promoter of the histone deacetylase gene SIRT1 as assessed by ChIP (Strub et al, 2011). Consistent with this, knockdown of MITF-M by siRNA reduces the levels of SIRT1 mRNA and protein expression in melanoma cell lines and patient biopsy samples, concurrent with a reduction in the levels of deacetylation of known SIRT1 targets such as TP53 (Ohanna et al, 2014).
MITF-M-dependent SIRT1 expression contributes to proliferation and suppression of senescence in melanoma cells (Ohanna et al, 2014; reviewed in Goding and Arnheiter, 2019). SIRT1 expression may also contribute to metabolic reprogramming through deacetylation of targets such as PPARGC1A and CBP/p300 (Bosch-Presegue and Vaquero, 2011; Canto et al, 2009; Bouras et al, 2005)
