Cytosolic calcium downstream of PIEZO1 and signaling through ATP:P2RY2 and Gq/11 activate the phosphorylation of PDPK1 (PDK1) on serine-241 (Jin et al. 2021). PDPK1 in vitro spontaneously auto-phosphorylates serine-241 (Gao and Harris 2006); however, which kinase acts in vivo is unknown. Full autophosphorylation of PDPK1 on serine-241 and threonine-513 occurs after PDPK1 has bound phosphatidylinositol-3,4,5-trisphosphate (PIP3) (Gao and Harris 2006); however, the activity of PI3K, a source of PIP3, is not required during activation of PDPK1 in response to shear flow (Jin et al. 2021)
