Autolytic cleavage of DAG1 pre-protein yields alpha- and beta-DAG1 which remain non-covalently associated. Alpha-DAG1 is extensively N- and O-glycosylated in the ER and the Golgi, modifications that are required for its binding to laminin in the extracellular matrix Beta-DAG1 is also N-glysocsylated and remains assoicated with alpha-DAG1 at its cysteine-rich domain. In the Golgi, DAG1 alpha and beta-chains associate with other transmembrane components of the dystrophin-glycoprotein complex (DGC) such as the sarcoglycans and sarcospanin (Herzog et al, 2004; Akhavan et al, 2008; Oppizzi et al, 2008; Bozzi et al, 2009; reviewed in Bhat et al, 2018; Gawor and Prószyński, 2018).
