Notch2 and phosphorylated Creb1 bind Gzmb promoter
In mouse CD8+ T-lymphocytes, Notch2 intracellular domain, NICD2, in complex with Rbpj binds the proximal Rbpj binding element (RBE) of Gzmb (granzyme B) promoter. Creb1, phosphorylated at serine S133, binds a CRE site located 40 bp downstream of the proximal RBE in Gzmb promoter. At the same time, Creb1 associates with NICD2:Rbpj complex and recruits transcriptional coactivator Ep300 (p300). This was demonstrated by chromatin immunoprecipitation (chIP) and gel mobility shift assays (Maekawa et al. 2008).
