Uhrf1 binds chromatin with hemimethylC
Uhrf1 (also known as Np95) preferentially binds hemi-methylated CG dinucleotides with its SRA domain (Arita et al. 2008, Hashimoto et al. 2008) and recruits Dnmt1 (Bostick et al. 2007). Uhrf1 flips the unmethylated cytosine base out of the DNA helix (Arita et al. 2008, Hashimoto et al. 2008). Uhrf1 also binds trimethylated lysine-9 of histone H3 (H3K9me3) which may direct Uhrf1 to transcriptionally silenced chromatin (Rottach et al. 2010).
