Shh binds Boc:Ptch1
Boc is a co-receptor for Hh signaling that binds both to Ptch1 and to Shh. Boc binding to Ptch1 is constitutive and not regulated by the interaction with Shh (Izzi et al, 2011). Binding of Boc to Shh is mediated by the third fibronectin domain of Boc, while binding of Boc to Ptch1 requires Boc fibronectin domains one and two (McLellan et al, 2008; Okada et al, 2006; Tenzen et al, 2006; Yao et al, 2006; Izzi et al, 2011). Different residues of Shh are required for interaction with Boc and Ptch1, suggesting the possibility of a ternary Shh:Boc:Ptch1 complex as depicted here, although this has not been explicitly demonstrated (Marigo et al, 1996; Stone et al, 1996; Chen and Struhl, 1996; Izzi et al, 2011; McLellan et al, 2008). Boc interacts with Ptch1 independently of Cdon and Gas1, two additional Hh coreceptors that are also required to activate Hh signaling (Izzi et al, 2011; Allen et al, 2011).
