Cxxc4 binds DVL to prevent AXIN binding
Cxxc4 was identified as Idax in a two-hybrid screen of a rat library with a human DVL1 bait. Idax forms a complex with DVL in vivo and competes with AXIN for DVL binding. Expression of Idax in L-cells abrogates the WNT3a-induced expression of a TCF-dependent reporter gene, supporting a role for Idax as a negative regulator of WNT target gene expression (Hino et al, 2001).
