Torcetrapib associates with a molecule of CETP and a spherical HDL particle to form a stable complex, thus trapping CETP and inhibiting CETP-mediated lipid transfer between HDL and LDL (Clark et al. 2006). Torcetrapib was a drug developed to treat hypercholesterolemia and prevent cardiovascular disease but its development was stopped when phase III studies showed excessive all-cause mortality in the treatment group (Howes & Kostner 2007).
